چکیده

Abstract WNT-medulloblastoma has an excellent prognosis, with overall survival rate of 90% among children receiving standard-of-care surgical resection, radiotherapy, and chemotherapy. While curative, this treatment is associated major, long-term, debilitating side effects. Therefore, it crucial we develop effective, less toxic therapies for these children. The Gilbertson lab previously shown that WNT-medulloblastomas are devoid a Blood-Brain-Barrier (BBB). This disruption allows large molecules such as monoclonal antibodies (mAbs) to penetrate WNT-medulloblastomas. To discover specific antigens mAb targeting, have first generated bulk single-cell RNA sequencing profiles the wild-type WNT-medulloblastoma-prone mossy-fibre progenitor lineages in lower rhombic lip, well human medulloblastoma patient derived orthotopic xenograft (PDOX) tumours. We next plasma membrane proteome hindbrain germinal zones Plasma Membrane Profiling (PMP) deploys isobaric chemical labelling approach -Tandem Mass Tags (TMT) - multiplexing up sixteen variables per mass spectrometry experiment ex vivo TMT-labelling primary cells. Using approach, mapped cell surface our models. Interrogation developmental tumour data unmasked prenatal brain WNT-medulloblastoma, but not expressed on critical postnatal tissues. Our workflow integrates multi-omic sets uncover targetable landscape cells cross-validated cohort histological samples both mouse translate dataset real benefit, generating high affinity validated cell-surface proteins assessment pre-clinical Through comparative analyses additionally identified non-WNT-specific targets thus aim expand identifying other more agressive CNS tumours metastases.

Proteomic profiling of high risk medulloblastoma reveals functional biology

Genomic characterization of medulloblastoma has improved molecular risk classification but struggles to define functional biological processes, particularly for the most aggressive subgroups. We present here a novel proteomic approach to this problem using a reference library of stable isotope labeled medulloblastoma-specific proteins as a spike-in standard for accurate quantification of the tu...

Particle in Cell-Monte Carlo Collisions of a Plasma Column Driven by Surface Wave Plasma Discharges

In this work, applicability of Particle in Cell-Monte Carlo Collisions (PIC-MCC) simulation method for better understanding of the plasma physical mechanisms and real important aspects of a plasma column driven by surface wave plasma discharges that is used in plasma antennas is examined. Via the implementation of geometry and physical parameters of the plasma column to an Object Oriented PIC-M...

Cell surface Thomsen-Friedenreich proteome profiling of metastatic prostate cancer cells reveals potential link with cancer stem cell-like phenotype

The tumor-associated Thomsen-Friedenreich glycoantigen (TF-Ag) plays an important role in hematogenous metastasis of multiple cancers. The LTQ Orbitrap LC-MS/MS mass spectrometry analysis of cell surface TF-Ag proteome of metastatic prostate cancer cells reveals that several cell surface glycoproteins expressing this carbohydrate antigen in prostate cancer (CD44, α2 integrin, β1 integrin, CD49f...

An unusual cell surface modification: a double plasma membrane.

The occurrence of an unusual double plasma membrane structure is reported; it has been studied in conventional thin sections, after lanthanum-impregnation and with freeze-fracturing. This modification of the plasmalemma is found where the luminal cell membrane (I membrane) of gut microvilli in the haematophagous insect, Rhodnius prolixus, is surrounded by a second, outer membrane (O membrane), ...
